The present invention provides methods of treating Alport syndrome in a subject by the administration of an agent that can blocks the activation of RAC1/CDC42 members of the rho family of small GTPases. Such agents include, but are not limited to, the endothelin receptor antagonist bosentan. Such administration prevents invasion of the glomerular capillary tufts by mesangial lamellipodial/filopodial processes, blocks mesangial process invasion abrogates the deposition of laminin 211 in the GBM, and prevents the activation of maladaptive expression of proteins known to contribute to glomerular disease progression.La présente invention concerne des procédés permettant de traiter le syndrome dAlport chez un sujet par ladministration dun agent qui peut bloquer lactivation des membres RAC1/CDC42 de la famille rho de petites GTPases. Lesdits agents comprennent, sans toutefois sy limiter, le bosentan antagoniste des récepteurs de lendothéline. Ladite administration prévient linvasion des floculus glomérulaires par des processus lamellipodiaux/filopodiaux mésangiaux, bloque linvasion des processus mésangiaux, abroge le dépôt de laminine 211 dans le GBM, et prévient lactivation de lexpression mal adaptée de protéines connues pour contribuer à la progression de la maladie glomérulaire.
