The present disclosure relates generally to compositions and methods for the treatment of cancer. In some embodiments, disclosed herein is a method for the induction of synthetic lethality by epigenetic therapy (ISLET) using a combination of at least one epigenetic compound and at least one chemotherapeutic agent. Also disclosed herein are screening methods for identifying compounds that induce cancer cell killing when combined with at least one epigenetic compound. Further provided herein is a method of enhancing the therapeutic effect of a chemotherapeutic agent on cancer, comprising administering to a subject having cancer an amount of an epigenetic compound effective to enhance the therapeutic effect of the chemotherapeutic agent on cancer.
