PURPOSE: A method for preparing chitosan microsphere is provided to produce chitosan microsphere having improved elasticity and flexibility. CONSTITUTION: A method or preparing chitosan microsphere comprises: a step of making chitosan solution； a step of dropping chitosan solution to upper layer of organic solvent and lower layer of alkali solution or cationic solution； and a step of cross-linking the microsphere. The organic solvent which is not soluble with water is petroleum ether or ethel ether. Dropping is performed using nitrogen gas. The cross-linking is performed by reacting with genipin.본 발명은 키토산을 사용하여 그 크기가 균일하고, 형태가 구형인 마이크로스피어를 제조하는 방법을 제공하며, 더욱 바람직하게는 크기가 균일하고, 구형일 뿐만 아니라, 팽윤성이 뛰어나고, 탄력성/유연성이 향상된 키토산 마이크로스피어(미세구)를 제조하는 방법을 제공한다.
