Compositions for modulating LAIR-1 and methods of use thereof are provided. For example, immunomodulators are provided that reduce LAIR-1 expression, ligand binding, cross-linking, negative signaling, or a combination thereof. Such agents can be used to provide an increased immune response to a subject in need thereof. Examples of agents include (i) soluble LAIR-2 polypeptide or fusion protein, (ii) soluble LAIR1 polypeptide or fusion protein, (iii) function blocking anti-LAIR-1 antibody, (iv) LAIR-1-positive cells Antibodies that are depleted, and (v) combinations thereof. Also provided are immunomodulatory agents that increase LAIR-1 expression, ligand binding, cross-linking, negative signaling, or a combination thereof. Such agents can be used to cause a reduction in immune response in a subject in need thereof. Examples of agents include (i) function-activating anti-LAIR-1 antibodies, (ii) function-blocking anti-LAIR-2 antibodies, and (iii) combinations thereof.
