Disclosed is a method for the detection of the presence of Mycobacterium, for example Mycobacterium bovis, in a biological sample, said method comprising the steps: providing a biological sample from an animal and determining in vitro the presence or absence of, or the presence or absence of an immune response to, each member of the group of antigens comprising: an Rv3616 antigen, an MPB70 antigen, an MPB83 antigen, and an ESAT 6 antigen wherein the identification of the presence of two or more of said antigens or of an immune response thereto is indicative of the presence of Mycobacterium in the biological sample. Further disclosed is a method of diagnosis of tuberculosis in a mammal, for example a reservoir mammal, the method comprising the steps: providing a biological sample from said animal and determining in vitro the presence or absence of, or the presence or absence of an immune response to, each member of the group of antigens comprising an Rv3616 antigen, an MPB70 antigen, and an MPB83 antigen, wherein the identification of the presence of one or more of said antigens of said group of antigens, or of an immune response thereto, is indicative of the presence of tuberculosis infection in the mammal.
