The present invention discusses the delivery of synergistic antibacterial agents to localized sites in the periodontal cavity for the treatment of periodontal disease by the aid of coated delivery device. Invention is related to optimized coated synthetic device for the treatment of periodontal infections. Therapeutic agents are mixed in a polymeric matrix, coated onto delivery device which is inserted into periodontal cavity, during which the drugs diffuses out of the coated device, providing targeted sustained release of drug(s). In the present invention, an attempt is made to develop a low-dose controlled-release delivery system for the treatment of periodontal infections. Synthetic device were taken as core material. The coating solution contained sustained release polymers and antibacterial drugs. The developed device is coated several times to maximize drug(s) loading. The coating composition was optimized on the basis of in vitro release studies and microbiological assays. For the present invention in vitro in vivo correlation is studied by especially designed continuous flow through apparatus and is found to be of level C correlation. The invention is further validated by microbiological parameters such as minimum inhibitory concentrations. The drug(s) coated device provided sustained effect up to a period of 168 hours. By optimizing coating solution concentration, release characteristics of the device is modified to provide sustained and targeted delivery over a period of seven days, duration of contact and antibacterial concentration is achieved which results in complete inhibition of all bacterial growth in dental cavity. The developed targeted drug(s) delivery sustained release device is studied along all in vivo periodontal parameters.
