The present invention relates to therapeutic ADCs comprising SN-38 conjugated to anti-Trop-2 antibodies or antigen-binding antibody fragments, and more particularly to sasituzumab gobinotecan. ADCs are administered to subjects with Trop-2 positive cancer that are resistant to, or have relapsed from, pretreatment with a checkpoint inhibitor. The treatment is effective in treating cancers that are resistant to checkpoint inhibitors.
