There is provided a method and composition for making and using a non-human animal and its animal wherein the non-human animal has a genome engineered into an immunoglobulin heavy chain variable region or comprising a recombinant diversity cluster, wherein the engineered or recombinant variant Clusters include the insertion of one or more coding sequences of a non-immunoglobulin polypeptide of interest. The non-human animal described herein expresses an antibody characterized by a complementarity determining region (CDR), particularly a CDR3 with diversity that induces binding to a particular antigen.
