The invention pertains to the use of a composition in the manufacture of a productor method for therapeutically improving, promoting, restoring or maintaining white matter integrity in a subject suffering from PKU and/or in a subject suffering from or at increased risk of impaired white matter integrity associated with PKU, said composition comprising therapeutic amounts of at least one ω-3 polyunsaturated fatty acid (LCPUFA) selected from the group consisting of docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA) and docosapentaenoic acid (DPA) and one of more of uridine, cytidine and/or an equivalent thereof, including salts, phosphates, acyl derivatives and/or esters.The invention also pertains to a composition for use in such use or method.Linvention concerne lutilisation dune composition dans lélaboration dune méthode de production permettant daméliorer, de favoriser, de restaurer ou de maintenir thérapeutiquement lintégrité de la substance blanche chez un sujet atteint de phénylcétonurie et/ou chez un sujet atteint ou présentant un risque accru de dégradation de lintégrité de la substance blanche associée à la phénylcétonurie, ladite composition comprenant des quantités thérapeutiques dau moins un acide gras polyinsaturé ω-3 (LCPUFA) choisi dans le groupe constitué par lacide docosahexaénoïque (DHA), lacide eicosapentaénoïque (EPA) et lacide docosapentaénoïque (DPA) et au moins lune de luridine, la cytidine et/ou un équivalent de celles-ci, comprenant des sels, des phosphates, des dérivés dacyle et/ou des esters.
