PROBLEM TO BE SOLVED: To provide a method of manufacturing a mold with which the needle tip of a protrusion pattern of a model is not damaged, and also to provide a method of manufacturing a pattern sheet using the mold.SOLUTION: The method of manufacturing a mold 68 includes the steps of: forming a silicone resin film 60 on a surface of a model 40 having a protrusion pattern 42； pressing a side of a base plate 64 with a separation sheet 66 in intimate contact therewith against a gap adjustment mechanism 52 and against the silicone resin film 60； heating and curing the silicone resin film 60 to form the mold 68； releasing the base plate 64； and releasing the separation sheet 66 from the mold 68 after releasing the mold 68 from the model 40. In the method, the gap adjustment mechanism 52 has a height at which a tip of the protrusion pattern 42 sticks into the separation sheet 66 when the base plate 66 is pressed against the gap adjustment mechanism 52.
