Provided is a composition comprising a fusion protein and a desired antigen,wherein the fusion protein comprisesa chaperone protein like heat shock protein 65 and a capsid protein like HBVcore antigen, which can produce immunogenicmacro-molecular structures as antigen carrier. Also provided are preparationmethods of the composition, which comprise purifyingrecombinant capsid proteins in the presence of chaotropic agents like urea orguanidine hydrochloride, and reassembling of thedenatured recombinant capsid proteins. The composition can be used fortherapeutic or prophylactic vaccination.
