Chimeric flaviviruses that are avirulent and immunogenic are provided. Thechimeric viruses are constructed to contain amino acid mutations in thenonstructural proteins of a flavivirus. Chimeric viruses containing theattenuation-mutated nonstructural genes of the virus are used as a backboneinto which the structural protein genes of a second flavivirus strain areinserted. These chimeric viruses elicit pronounced immunogenicity yet lack theaccompanying clinical symptoms of viral disease. The attenuated chimericviruses are effective as immunogens or vaccines and may be combined in apharmaceutical composition to confer simultaneous immunity against severalstrains of pathogenic flaviviruses.
