The present disclosure provides methods of treating a disease or delivering a therapeutic agent to a mammal comprising administering to the mammal's cisterna magna and/or ventricle an rAAV particle containing a vector comprising a nucleic acid encoding a therapeutic protein inserted between a pair of AAV inverted terminal repeats in a manner such that cells with access to the cerebrospinal fluid (CSF) express the therapeutic agent and in certain embodiments secretes the therapeutic agent into the CSF for distribution to the brain.La présente invention concerne des procédés de traitement d'une maladie ou de distribution d'un agent thérapeutique à un mammifère, consistant à administrer à la citerne cérébello-médullaire et/ou au ventricule du mammifère une particule de rAAV contenant un vecteur comprenant un acide nucléique codant pour une protéine thérapeutique inséré entre une paire de répétitions terminales inversées d'AAV, de telle manière que les cellules ayant accès au liquide céphalo-rachidien (LCR) expriment l'agent thérapeutique et dans certains modes de réalisation sécrètent l'agent thérapeutique dans le LCR pour le distribuer au cerveau.
