A range of therapeutic mRNA molecules that can be expressed to provide a target polypeptide or protein. The RNA molecule can contain one or more 5-methoxyuridine and 5-methylcytidine. Further provided is a DNA template that can be transcribed to provide the target mRNA and can have altered nucleotides such as reduced deoxyadenosine. The invention also provides processes for making therapeutic mRNA molecules.
