Relating to the preparation of a composition including a drug such as pemetrexed which is easily degraded through oxidation, disclosed are a composition having increased stability by removing oxygen during the process of preparation of and to a method of preparing same. The preparation method is made easier by freezing and degassing in a sealed chamber, and therefore the preparation method can provide a composition having remarkably increased stability of an oxidation-labile drug.
