This invention relates to a transgenic animal model for testing immunogenicity and protective efficacy of human vaccines and the method for generating such a multi-transgenic animal. This invention also relates to methods for screening compositions for human vaccine development. More specifically, the present invention relates to a mouse model capable of expressing human leukocyte antigens DR4 and A2, and/or human costimulatory molecules (CD80) which upon infusion of human HLA-matched hematopoietic stem cells develop a functional human immune system able to respond to vaccination with human vaccines. The invention also relates to method of producing human antibodies specific for a desired antigen using the transgenic mouse.Cette invention concerne un modèle animal transgénique pour le test de limmunogénicité et lefficacité de protection de vaccins humains et le procédé de génération dun tel animal multi- transgénique. Cette invention concerne également des procédés de criblage de compositions pour le développement de vaccins humains. Plus particulièrement, la présente invention concerne un modèle murin apte à exprimer des antigènes DR4 et A2 de leucocytes humains et/ou des molécules costimulatrices humaines (CD80), qui au cours de la perfusion de cellules souches hématopoïétiques humaines correspondant à un HLA développent un système immunitaire humain fonctionnel apte à répondre à la vaccination par des vaccins humains. Linvention concerne également un procédé de production danticorps humains spécifiques dun antigène souhaité à laide de la souris transgénique.
