PROBLEM TO BE SOLVED: To provide an endoscope system capable of acquiring biological numeric information of high objectivity without depending upon an illumination condition nor a photographing condition, and the operation method thereof.SOLUTION: A first still picture acquisition instruction is generated by a depression operation of a freeze button 13b. The light emission amount of illumination light emitted during the first still picture acquisition instruction is stored in a light emission amount storage part 24. The illumination light is emitted on the basis of the light emission amount stored in the light emission amount storage part 24. Brightness information is calculated from an image signal obtained by imaging an observation object.
