A genetically engineered mouse characterized by one or more symptoms or signs associated with non-familial late-onset Alzheimer's disease with expression of human APOE4p and mouse Trem2p, the mouse genome being 1) driven by a promoter A DNA sequence encoding a human APOE4 protein (APOE4p) operably linked, and 2) operably linked to a promoter, p. Provided is a genetically modified mouse comprising a DNA sequence encoding a mouse Trem2 protein having the R47H mutation (Trem2p), whereby the mouse expresses human APOE4p and mouse Trem2p.
