The present invention provides a method for separating and purifying high-purity vancomycin hydrochloride, which comprises (1) obtaining a vancomycin hydrochloride solution by ion-exchange chromatography, and further desalting and concentrating the solution by nanofiltration. And (2) first adjusting the concentrated solution with a hydrochloric acid solution, then passing the prepared concentrated solution through a reverse phase chromatography column and performing column chromatography, and (3) chromatography of vancomycin B. Collecting the liquid to obtain a mixed chromatographic liquid; (4) adjusting the mixed chromatographic liquid with hydrochloric acid and then concentrating with nanofiltration to remove the solvent and salt to obtain a concentrated liquid; (5) The concentrated liquid obtained in step (4) is dehydrated and dried, or is subjected to solvent crystallization or crystallization by salting out and chromatographed.
