The present invention provides a method for separating and purifying high-purity vancomycin hydrochloride, which comprises (1) obtaining a vancomycin hydrochloride solution by ion-exchange chromatography, and further desalting and concentrating the solution by nanofiltration. And (2) first adjusting the concentrated solution with a hydrochloric acid solution, then passing the prepared concentrated solution through a reverse phase chromatography column and performing column chromatography, and (3) chromatography of vancomycin B.
