The present invention relates to a multilayer cell sheet of neural crest stem cells and a method for preparing the same and, more specifically, to a method of preparing a multilayer cell sheet through a three-dimensional single culture process by introducing neural crest stem cells derived from peripheral nerves into a hydrogel using a biodegradable natural polymer hydrogel as a three-dimensional support. According to the present invention, the multilayered cell sheet does not require special equipment to manufacture, is easy to handle due to excellent physical properties thereof, increases the survival rate of cells after transplantation since various growth and protective factors and extracellular matrix are sufficiently accumulated between cells, and also facilitates nutrient migration since the thickness thereof is thin due to cell-induced contraction. Therefore, the multilayered cell sheet of neural crest stem cells is expected to be useful as a therapeutic agent for spinal cord injury or brain injury diseases.본 발명은 신경능선줄기세포의 다층세포시트 및 이의 제조방법에 관한 것으로서, 생분해성 천연고분자 하이드로젤을 3차원 지지체로 이용하여, 말초신경 유래 신경능선줄기세포를 하이드로젤 내로 함입시켜 3차원 단일 배양과정을 통하여 다층세포시트를 제조하는 방법을 제공하고자 한다. 본 발명의 다층 세포 시트는 제조하는데 특수한 기구가 필요하지 않으며, 물리적 특성이 좋아 다루기도 쉬우며, 세포 사이에 각종 성장 및 보호 인자 와 세포외 기질이 충분히 축적되어 이식 후 세포의 생존율을 높이며, 세포에 의해 유발된 수축에 의해 두께 또한 얇아 영양분 이동 또한 용이하다. 따라서, 상기 신경능선줄기세포의 다층세포시트는 척수 손상 질환 또는 뇌 손상 질환 치료제로 유용하게 활용될 수 있을 것으로 예상된다.
