The present invention provides ligand drug conjugates and drug-linker compounds comprising methylene carbamate units. The present invention provides, inter alia, ligand drug conjugates, where the ligand drug conjugate comprises a self-destructing assembly unit having a methylene carbamate unit for conjugating the drug to the targeting ligand. Methods and methods used, and intermediates thereof are provided.
