PROBLEM TO BE SOLVED: To provide a ligand drug conjugate containing a methylene carbamate unit and a drug-linker compound. The present invention relates, among other things, to ligand drug conjugates, wherein the ligand drug conjugate comprises a self-destructive assembly unit having a methylene carbamate unit for binding a drug to a targeting ligand. Methods for preparing and using them, as well as intermediates thereof are provided. The ligand drug conjugates of the invention are stable in circulation, but can result in cell death when the free drug is released from the conjugate in the vicinity of or within the tumor cells.
