A holding device disposed at a bio-detecting instrument, which has a plurality of sensing terminals, to hold a test strip. The holding device has a first case, a second case, an exiting mechanism and an elastic member. The second case covers to the first case, and a space is formed between them. The sensing terminals are disposed at the space. The test strip movably enters to the space from a first opening to touch the sensing terminals. The exiting mechanism has a pushing rod, which can movably enter or exit the space from a second opening, and a first stopper. The elastic member is disposed between the second case and a first stopper. When pushing the exiting mechanism to an exiting position, the elastic member is compressed, and the pushing rod enters to the space to push out the sample without touching the sensing terminals.
