The present invention provides methods of site-specific labeling of antibodies, using proteins having 4′-phosphopantetheinyl transferase activity that catalyze post-translational modification of peptide sequences (“peptide tags”) incorporated into one or more specific sites of an antibody of interest. Enzymatic labeling enables quantitative and irreversible covalent modification of a specific serine residue within the peptide tags incorporated into the antibody, and thus creates desirable antibody conjugates.본 발명은 관심 항체의 하나 이상의 특이적 부위 내로 혼입된 펩티드 서열 ("펩티드 태그")의 번역후 변형을 촉매하는 4'-포스포판테테이닐 트랜스퍼라제 활성을 갖는 단백질을 사용하는, 항체의 부위-특이적 표지 방법을 제공한다. 효소적 표지는 항체 내로 혼입된 펩티드 태그 내의 특이적인 세린 잔기의 정량적이고 비가역적인 공유결합 변형을 가능하게 하고, 따라서 바람직한 항체 접합체를 생성시킨다.
