The present invention relates to a method and submerged culture medium for preparing high ergothioneine Pleurotus citrinopileatus mycelia. According to the method of present invention, an exemplary optimal P. citrinopileatus submerged culture conditions are 25℃, 22 days and 5% incubation rate with carbon and nitrogen sources being 2% glucose and 0.5% yeast extract and add amino acid complex solution (8 mM cysteine, 4 mM histidine and 0.5 mM methionine) after 7 days, the content of ergothioneine from P. citrinopileatus mycelia was 14.57 mg/g dw.本發明係關於一種製備高含量麥角硫因之珊瑚菇菌絲體液態培養方法及培養液。根據本發明方法,例舉之最適的珊瑚菇菌絲體深層培養條件為:溫度25ºC,接種量5%,以2%葡萄糖為碳源,以0.5%酵母抽出物作為氮源,並於培養第7天時添加綜合胺基酸液(半胱胺酸8mM、組胺酸4mM及甲硫胺酸0.5mM),可得菌絲體培養物之麥角硫因含量為14.57mg/g dw。
