The present invention relates to an insulin secretion peptide derivative in which the charge on the N-terminus is modified and a pharmaceutical composition comprising the same, specifically, is characterized in that, in the insulin secretion peptide derivative, the positive charge on the N-terminus of an insulin secretion peptide is changed to a neutral charge or net negative charge in a neutral pH. The insulin secretion peptide derivative according to the present invention exhibits higher insulin secretion ability and blood glucose-reducing ability than natural insulin secretion peptides while maintaining the stability in the bloodstream because dissociation from a GLP-1 receptor rapidly progresses due to the change in the charge on the N-terminus, and thus is very useful in treating Type 2 diabetes.
