Die Erfindung betrifft einen mitochondrialen Expressionsvektor umfassend ein zu exprimierendes Gen und/oder einen Selektionsmarker, und einen mitochondrialen Promotorbereich, sowie ein Verfahren zum Einbringen einer zu exprimierenden DNA in Mitochondrien von Säugerzellen, wobei das Verfahren die Schritte umfasst:(i) Konstruktion eines mitochondrialen Expressionsvektors oder eines mitochondrialen Genoms,(ii) reversible Induktion von Megamitochondrien und(iii) Transfektion der Megamitochondrien mittels einer physikalischen Transfektionsmethode.Mitochondrial expression vector comprising a gene to be expressed and/or a selection marker, and a mitochondrial promoter region is new. Independent claims are included for: (1) inserting DNA to be expressed in the mitochondria of mammalian cells, comprising constructing a mitochondrial expression vector or a mitochondrial genome, performing reversible induction of mega-mitochondria and transfecting the mega mitochondria by a physical transfection method； and (2) reversible induction of mega mitochondria of mammalian cells by acidification of mammalian cells culture medium by addition of sodium acetate or lactic acid.
