A method of cultivating Cordyceps militaris according to the present invention comprises: a step of inoculating a liquid medium, a solid medium, or an insect with Cordyceps militaris or Cordyceps nutans to obtain mycelia； and a step of culturing the obtained mycelia in a solid medium or an insect to form a fruit body. The solid medium forming a fruit body is composed of corns and a liquid medium, or is composed of rice, silkworm pupae, and a liquid medium； and has a water content of 50-70%, a relative humidity of 80-95%, an ambient temperature of 15-25°C； and has a luminous intensity of 700-1,200 lux. The liquid medium yielding mycelia is composed of MgSO_4·7H_2O, KH_2PO_4, K_2HPO_4, a yeast extract, peptone, and dextrose, or is composed of a yeast extract, dextrose, EBIOS, and Hyponex. The solid medium yielding mycelia is composed of a liquid medium and at least one selected from rice, potato, corn, and silkworm pupae.본 발명에 따른 동충하초 배양 방법은 액체 배지, 고형 배지 또는 충체에 코디셉스 밀리타리스 또는 코디셉스 누탄스균을 접종하여 균사체를 얻는 단계； 및 상기 얻어진 균사체를 고형 배지 또는 충체에서 배양하여 자실체를 형성시키는 단계를 포함하고, 상기 자실체를 형성하는 고형 배지는 옥수수와 액체 배지로 이루어지거나, 쌀, 누에고치 번데기 및 액체 배지로 이루어지고, 수분 함량 50 내지 70 %, 상대습도 80 내지 95 %, 분위기 온도 15 내지 25 ℃ 및 광도 700 내지 1200 럭스가 되고, 상기 균사체를 얻는 액체 배지는 MgSO₄·7H₂O, KH₂PO₄, K₂HPO₄, 효모 추출물, 펩톤 및 덱스트로오스로 이루어지거나 또는 효모 추출물, 덱스트로오스, 에비오스 및 하이포넥스로 이루어지는 것을 특징으로 하고, 상기 균사체를 얻는 고형 배지는 쌀, 감자, 옥수수 및 누에고치 번데기로 이루어진 군으로부터 선택된 적어도 하나 및 액체 배지로 이루어진다.
