The present invention provides compositions and methods for delivering a therapeutic agent across a barrier. Such compositions comprise a therapeutic agent (e.g., an antimicrobial agent, an antibiotic, or an anesthetic agent), a penetration enhancer to increase the flux of the therapeutic agent across the barrier, and a matrix former. The matrix formers form gels at suitable gelation temperatures and rheological properties for use in drug delivery, and in some cases, such gelation temperature and rheological properties are not significantly changed from those of compositions without penetration enhancers. The present invention also provides matrix formers and compositions thereof. Such compositions are particularly useful for the treatment of otitis media.
