The present invention relates to a method for efficiently cultivating tetraploid watermelon varieties by chemical treatment in a seedling obtained by in vitro germination of watermelon seeds. The method according to the present invention comprises the following steps of: (a) germinating sterilized watermelon seeds in an MS culture medium and forming a seedling； (b) treating growing points of the seedling in the step (a) with chemicals of colchicine and 2,6-dinitroaniline； (c) obtaining fragments by cutting cotyledons and growing points of the seedling treated with chemicals in the step (b), and planting the obtained fragments in the MS culture medium to induce shoots； (d) growing the shoots in the step (c) in the MS culture medium； (e) inducing the rooting of the grown shoots in the step (d) in the MS culture medium； (f) purifying the rooted watermelon plant bodies in the step (e) to the soil environment； and (g) transplanting the purified watermelon plant bodies in the step (f) into a flowerpot and growing the same.본 발명은 수박 종자를 기내 발아시켜 확보된 유묘에 약품처리에 의한 효율적인 4배체(tetraploid) 유기방법에 관한 것으로, 그 구성은,a) 살균처리된 수박종자를 MS배지에서 발아 및 유묘를 형성시키는 단계； b) 상기 a) 단계의 유묘 생장점에 콜히친과 2,6-dinitroaniline의 약제를 처리하는 단계； c) 상기 b) 단계의 약제처리된 유묘의 자엽과 생장점을 절취하여 절편을 수득하고 그 수득 된 절편을 MS 배지에 치상하여 신초를 유도하는 단계； d) 상기 c) 단계의 신초를 MS 배지에서 신장시키는 단계； e) 상기 d) 단계의 신장 된 신초를 MS 배지에서 발근 유도하는 단계； f) 상기 e) 단계의 발근 된 수박 식물체를 토양환경에 순화시키는 단계； g) 상기 f) 단계의 순화된 수박 식물체를 화분에 이식하여 생육시키는 단계； 로 이루어진다.
