Provided are methods of treatment for particular cancers and determining response to therapy with an anti-cancer agent that inhibits the activity of an enzyme that mediates repair of a DNA strand break, said methods including determining a property of an NPM1 nucleic acid or protein, such as an expression level, a mutational status and/or a subcellular location thereof.
