The present invention relates to the field of biotechnology, more particularly to a mutant polypeptide of a TGF beta molecule having a first sequence homologous to a sequence of human TGF beta. These mutant proteins lose the ability to interact with ALK5 but retain the ability to interact with other receptors (TβRII and TβRIII) that are part of the receptor complex. By adopting ALK5 in the receptor complex, they have antagonistic properties to the signaling of all natural mutants of the TGFβ ligand and a considerable immunoregulatory effect. The present invention relates to a pharmaceutical composition comprising the disclosed polypeptide or a fusion protein as an active ingredient, and to their use in diseases such as cancer, fibrosis, and chronic infectious diseases in consideration of the immune system controlling effect thereof.본 발명은 생명공학 분야에 관한 것으로, 보다 특별하게는 인간 TGFβ의 서열과 상동성이 높은 일차 서열을 가진 TGFβ 분자의 돌연변이 폴리펩티드에 관한 것이다. 이들 돌연변이 단백질은 ALK5와 상호작용하는 능력을 상실하지만 수용체 복합체의 일부인 다른 수용체(TβRII와 TβRIII)와 상호작용하는 능력을 유지한다. 이들은 수용체 복합체에 ALK5를 채용함에 따라 TGFβ 리간드의 모든 자연 변이체의 신호전달에 길항하는 성질과 상당한 면역조절 효과를 갖고 있다. 본 발명은 개시되어 있는 폴리펩티드 또는 융합 단백질을 유효 성분으로서 포함하는 약제학적 조성물 및 암, 섬유증을 동반한 질환과 만성 감염성 질환과 같은 질환에서 이들의 면역계 조절 효과를 고려한 이들의 용도에 관한 것이다.
