PROBLEM TO BE SOLVED: To provide a method for engineering an immunoglobulin containing at least one modification in a structural loop region of the immunoglobulin and determining the binding of the immunoglobulin to an epitope of an antigen, wherein the unmodified immunoglobulin does not significantly bind to the epitope.SOLUTION: The method includes steps of: providing a nucleic acid encoding an immunoglobulin containing at least one structural loop region modifying at least one nucleotide residue of at least one of the structural loop regions transferring the modified nucleic acid in an expression system expressing the modified immunoglobulin contacting the expressed modified immunoglobulin with an epitope and determining whether or not the modified immunoglobulin binds to the epitope.
