The disclosed bead vectors are directed to invade cells of monocyte origin and cause expression of antibodies specific for the extracellular domain of PD-1, anti-CTLA4 antibody, or checkpoint protein. The bead vector can include a nucleic acid component, a lysosome escape component and a bead particle that can be phagocytosed. The disclosed vectors are useful in a variety of methods for cancer therapy, treatment, and prevention. Due to the ability of monocyte cells to target tumors, the disclosed vectors are particularly well suited for use in anti-tumor applications and for targeting target gene expression in tumor-associated macrophages.
