The present invention relates to a polymer delivery system for RNAi therapies that includes diseases where siRNA may be effective. In particular the invention relates to polymeric structures that self assemble into nano particles for siRNA delivery. Such polymers include a block copolymer comprising a cationic polymer capable of binding to siRNA, a p H mediator capable of disrupting endosomes and a polymer capable of binding with cell membrane thereby to release the block copolymer into the cytoplasm and wherein the cationic polymer is capable of degradation thereby to deliver siRNA.La présente invention concerne un système polymère de libération pour des thérapies ARNi pour des maladies contre lesquelles lARNsi peut être efficace. En particulier, linvention concerne des structures polymères qui sauto-assemblent en nanoparticules pour la libération dARNsi. De tels polymères comprennent un copolymère séquencé contenant un polymère cationique capable de lier ARNsi, un médiateur de pH capable de déstabiliser les endosomes et un polymère capable de lier la membrane cellulaire pour ainsi libérer le copolymère séquencé dans le cytoplasme, le polymère cationique pouvant se dégrader et ainsi libérer lARNsi.
