In the present invention, the present invention relates to a novel method for preparing talirelin and an intermediate thereof, and more specifically, using Fmoc (fluorenylmethoxycarbonyl)-trityl-histidine as a starting material and in situ. By preparing talirelin through a process of preparing histidyl-trityl-prolineamide, which is an intermediate of talthyrlin as a reaction, it is possible to manufacture talirelin with high yield and high purity in a short time compared to the conventional technology, which is suitable for industrial mass production. Disclosed are a method for producing and a method for producing histidyl-trityl-prolineamide.본 발명에서는 탈티렐린 및 이의 중간체의 신규한 제조방법에 관한 것으로, 더 상세하게는 Fmoc(플루오레닐메톡시카르보닐)-트리틸(trityl)-히스티딘을 출발물질로 사용하고 인시츄(In situ) 반응으로 탈티렐린 중간체인 히스티딜-트리틸-프롤린아미드를 제조하는 공정을 거쳐 탈티렐린을 제조함으로써, 종래의 기술 대비하여 단시간에 고수율·고순도로 탈티렐린을 제조할 수 있어 산업적 대량 생산에 적합한 제조방법 및 이를 위한 히스티딜-트리틸-프롤린아미드의 제조방법이 개시된다.
