PROBLEM TO BE SOLVED: To provide novel polysaccharide derivatives of insulin and production methods thereof.SOLUTION: Provided are a pharmaceutical composition and a preparation method thereof, where the composition comprises a population of polysaccharide derivatives of a protein, the protein is insulin or an insulin-like protein, and the polysaccharide is anionic and comprising between 2 and 125 saccharide units, the population substantially comprises only N-terminal derivatives of the protein. Usually, the polysaccharide is PSA, the anionic polysaccharide has reactive aldehyde residues reactive with insulin or an insulin-like protein, the derivatization reaction is carried out under reducing conditions.
