PROCÉDÉS ET COMPOSITIONS POUR LA MULTIPLICATION DE CELLULES SOUCHES ET/OU PROGÉNITRICES HÉMATOPOÏÉTIQUES À L'AIDE D'UN INHIBITEUR DE CYTOCHROME P450 1B1 (CYP1B1) OU D'UN ACTIVATEUR DE MUSASHI-2 (MSI2)
A method of increasing the self-renewal and/or expansion of hematopoietic stem and/or progenitor cells (HSPCs) is described. Inhibiting the activity and/or expression of cytochrome P450 1B1 (CYP1B1) and/or increasing the expression or activity of Musashi-2 (MSI2) increases the expansion of HSPCs. The HSPCs may be cultured in the presence of a CYP1B1 inhibitor and/or a MSI2 activator. Optionally, the cells may be expanded ex vivo and transplanted into a subject in need thereof.L'invention concerne un procédé d'augmentation de l'auto-renouvellement et/ou de la multiplication de cellules souches et/ou progénitrices hématopoïétiques (HSPC). L'inhibition de l'activité et/ou de l'expression du cytochrome P450 1B1 (CYP1B1) et/ou l'augmentation de l'expression ou de l'activité de Musashi-2 (MSI2) augmente la multiplication des HSPC. Les HSPC peuvent être cultivées en présence d'un inhibiteur de CYP1B1 et/ou d'un activateur de MSI2. Éventuellement, les cellules peuvent être multipliées ex vivo et transplantées chez un sujet en ayant besoin.
