The present invention relates to a method for detecting a fluorescent species (P) that is reversibly photoswitchable at a high frequency, and more specifically to a method for detecting at least one reversibly photoswitchable fluorescent species, comprising a step of illuminating a sample containing a reversibly photoswitchable fluorescent species with a first illumination light (FEX1), of wavelength λ1, and periodically modulated at a pulsation ω and with a second illumination light (FEX2), of wavelength λ2, different from λ1, periodically modulated at a pulsation ω, the second illumination light being modulated in antiphase with respect to said first illumination light.La présente invention concerne un procédé de détection d'une espèce fluorescente réversiblement photocommutable (P) à haute fréquence, et plus précisément un procédé de détection d'au moins une espèce fluorescente réversiblement photocommutable, comportant une étape consistant à éclairer un échantillon contenant une espèce fluorescente réversiblement photocommutable avec une première lumière d'éclairage (FEX1), de longueur d'onde λ1, et modulée périodiquement à une pulsation ω et avec une seconde lumière d'éclairage (FEX2) de longueur d'onde λ2 différente de λ1, modulée périodiquement à la pulsation ω, la seconde lumière d'éclairage étant modulée en antiphase par rapport à ladite première lumière d'éclairage.
