This invention pertains to pharmaceutical composition comprising a plurality of multi-layered beads having an oxycodone layer and a sequestering subunit comprising a naltrexone and a blocking agent, in particular pharmaceutical compositions comprising a higher level of naltrexone, and related compositions and methods of use, such as in the prevention of abuse of a therapeutic agent.
