Disclosed herein are spiro-oxindole-based compounds of formula (I), compositions, and methods in the field of medicinal chemistry, wherein the variables shown in formula (I) are as defined in the specification. The compounds and compositions provided herein relate to spiro oxindoles which function as antagonists of the interaction between p53 and MDM2, and their use as therapeutics for the treatment of hyperproliferative disease, particularly cancer selected from the group consisting of melanoma, lung cancer, sarcoma, colon cancer, prostate cancer, choriocarcinoma, breast cancer, retinoblastoma, stomach carcinoma, acute myeloid leukemia, lymph multiple myeloma, and leukemia.
