This invention provides novel amine compounds of the following general formula (I) which exhibit excellent property in immunosuppressive action, anti-rejection activity, etc. and less side effects in bradycardia and others, or their pharmaceutically acceptable acid adduct salts； hydrates or solvates thereof； and pharmaceutical compositions comprising these compounds and pharmaceutically acceptable carriers,(wherein R is a hydrogen atom or P(=O)(OH)2； X is an oxygen atom or sulfur atom； Y is CH2CH2or CH=CH； R1is a halogenated C1-C4alkyl or cyano； R2is an optionally hydroxylated or halogenated C1-C4alkyl； R3and R4are same or different, each represents a hydrogenatom or C1-C4alkyl； n represents 5 to 8).本發明係有關於免疫抑制作用、排斥反應抑制作用等優越，減輕徐脈(bradycardia)等副作用之下述通式(I)表示之新穎胺化合物、或其製藥上可接受之酸加成鹽、該等之水合物或溶劑合物及含有該等化合物及製藥上可接受之載體之醫藥組成物。(式中，R表示氫原子或P(＝O)(OH)2，X表示氧原子或硫原子，Y表示CH2CH2或CH＝CH，R1表示以鹵素原子取代之碳原子數1至4之烷基或氰基，R2表示可以羥基取代或可以鹵素原子取代之碳原子數1至4之烷基，R3及R4可相同亦可不同，各表示氫原子或碳原子數1至4之烷基，n表示5至8)。
