This invention relates to a closed system medicine preparing and giving device, comprising a pushing barrel and a connection seat. The pushing barrel comprises an outer barrel, an inner barrel, and a pushing rod, wherein the inner barrel is penetratingly disposed in the outer barrel the pushing rod is movably disposed in the inner barrel the outer barrel comprises an air inlet tube and the inner barrel comprises a medicine inlet tube. The connection seat comprises a connection cover, two hole expansion needles, an elastic valve, and a water stopping member, wherein two hole expansion needles is covered in the elastic valve the elastic valve is disposed in the connection cover and the water stopping member is disposed in one of the hole expansion needles. When a user inserts the pushing barrel on the connection seat, the air inlet tube and the medicine inlet tube will compress the elastic valve to make those two hole expansion needles penetrating the elastic valve to respectively enter the medicine inlet tube and the air inlet tube.
