Disclosed are antiproliferative manoalide compounds extracted from the sponge Luffariella sp., their pharamaceutical composition, preparation method and identification method for the configuration. The compound has a structural formula (I): where R is CH3, the configuration combination for C-24 and C-25 is 24R,25R, 24R,25S or 24S,25S. The compound (I) is obtained by extracting Luffariella sp. with ethyl acetate (EtOAc), concentrating the EtOAc extract to obtain a residue, chromatographing the residue to yield fractions, and separating the compound (I) from fractions by the column chromatography on silica gel and reverse-phase HPLC.
