The present invention relates to nucleic acid and amino acid sequences of variants of human neprilysin with altered substrate specificity relative to wild-type human neprilysin and use of such variants in pharmaceutical compositions. In particular, the present invention relates to neprilysin variant polypeptides with increased specificity for cleavage of amyloid beta (Aj3) peptides compared to wild-type neprilysin. The invention also relates to fusion proteins comprising such neprilysin variant molecules.
