The present invention relates to nucleic acid and amino acid sequences of variants of human neprilysin with altered substrate specificity relative to wild-type human neprilysin and use of such variants in pharmaceutical compositions. In particular, the present invention relates to neprilysin variant polypeptides with increased specificity for cleavage of amyloid beta (Aj3) peptides compared to wild-type neprilysin. The invention also relates to fusion proteins comprising such neprilysin variant molecules. Polypcptidcs comprising ncprilysin variants may be used in the treatment of diseases associated with accumulation of amyloid beta, in particular Alzheimer's disease.La presente invención se refiere a polipéptidos que comprenden variantes de proteasa de neprilisina humano tipo silvestre que tiene una especificidad y/o actividad alterada.
