Checkpoint receptors and their cognate ligands are frequently targeted by B and T cells in autoimmune disorders, and their adaptive immune response may contribute significantly to the underlying immunopathology. Described herein are novel techniques for clonal exclusion of autoreactive B cells that target checkpoint receptors and their ligands. One embodiment of this technique is a checkpoint receptor or ligand extracellular domain molecule chimera with an effector domain capable of inducing B cell apoptosis, necrosis, and/or tolerization/anergization. Yes, this technique is referred to herein as PantId (polyclonal anti-idiotype). In other embodiments, the technique also includes effector molecule chimeras with immunomodulatory cytokines. Novel apoptotic effectors are also described. Methods of identifying checkpoint receptor/ligand autoreactive B cell responses, constructing PantIds, and their in vitro and in vivo applications are also described.
