The present invention relates to engineered a cytolytic immune cell comprising: i) a releasable protein which comprises a polypeptide of interest (POI) and a first interaction domain； and ii) a retention protein which is retained within an intracellular compartment of the cell and comprises a second interaction domain which binds to the first protein interaction domain, wherein binding between the first protein interaction domain and second protein interaction domain is disrupted by the presence of an agent, such that in the absence of the agent, the first protein interaction domain and second protein interaction domain bind and result in retention of the POI within an intracellular compartment； whereas in the presence of the agent, the first protein interaction domain and second protein interaction do not bind and the POI is released from the intracellular compartment and expressed at the cell surface or secreted by the cell.La présente invention concerne une cellule immunitaire cytolytique modifiée comprenant : i) une protéine libérable qui comprend un polypeptide d'intérêt (POI) et un premier domaine d'interaction； et ii) une protéine de rétention qui est retenue dans un compartiment intracellulaire de la cellule et comprend un second domaine d'interaction qui se lie au premier domaine d'interaction de protéine, la liaison entre le premier domaine d'interaction de protéine et le second domaine d'interaction de protéine étant rompue par la présence d'un agent, de telle sorte qu'en l'absence de cet agent, le premier domaine d'interaction de protéine et le second domaine d'interaction de protéine se lient et conduisent à la rétention du POI à l'intérieur d'un compartiment intracellulaire； tandis qu'en présence de l'agent, le premier domaine d'interaction de protéine et le second domaine d'interaction de protéine ne se lient pas et le POI est libéré du compartiment intracellulaire et exprimé au niveau de la surface cellulaire ou sécrété par la cellule.
