An aspect of the present invention includes a polyalkylene oxide-asparaginase composition. In one example, the composition is a freeze-dried storage stable composition. In certain instances, the lyophilized composition includes one or more of a buffer, a salt, and a sugar. Embodiments of the invention further include a method of making the composition. The composition finds use in a variety of applications such as, for example, treatment of a tumor condition in a subject.
