The present disclosure relates generally to the field of transdermal permeant delivery and more specifically to devices, systems and methods for using the same. More specifically the disclosure relates to patch for delivering a permeant through a biological membrane of a subject comprising: a) a matrix; b) at least one hydrophilic permeant disposed within the matrix, wherein at least a portion of the hydrophilic permeant can dissolve in biological moisture received from the subject; c) at least one permeability enhancer disposed within the matrix; and at least one of the following d1) and d2): d1) at least one water soluble polymer disposed within the matrix, wherein the water soluble polymer is at least one selected from the group consisting of: polyethylene glycol, polyvinyl alcohol, and polyvinylpyrrolidone; and d2) at least one solubility control agent selected from the group consisting of agents that selectively control pH of a solution relative to the isoelectric point of the hydrophilic permeant, ionic strength control agents, and salting-out agents, wherein when the patch does not comprise the at least one water soluble polymer, the hydrophilic permeant is at least one bioactive agent selected from the group consisting of peptides, polypeptides, proteins, oligonucleotides, nucleic acids, polysaccharides, and undissolved anhydrous hydrophilic salts excluding hydromorphone HCl. The disclosure also relates to a system comprising the above that additionally comprises a porator.
