PROBLEM TO BE SOLVED: To provide a primer set for detecting a DNA marker linked to a quantitative character gene locus relating to the fat content in a corn seed and to provide the use of the primer set. SOLUTION: The invention provides a primer set for detecting a DNA marker linked to a QTL located on the first, fourth and sixth chromosomes participating in the fat content of a corn seed, a method for discriminating the genotype of the fat-content relating QTL by performing PCR with corn genome DNA using the primer set and discriminating by the difference of the length of the amplified DNA fragment, and a method for preparing a high fat-content lineage by selecting a body having a fat-content relating QTL derived from a high fat-content lineage and preparing a lineage having high fat content in the seed in the posterior lineages.
